The residue was added with 20 ml of 10% hydrochloric acid solution, dissolved by micro-heating, hydrolyzed in a water bath for 2 hours, taken out, allowed to cool, filtered, and the precipitate was washed with water until neutral. The content of saponins in this product was determined by thin-layer scanning method. As a quantitative analysis, the stability, precision, reproducibility and recovery rate test proved that the method is accurate and reliable, and can play a role in controlling the quality of this product.

     

    Spectrophotometry

    Taking far-offic acid as the reference substance, vanillin-perchloric acid was used as the color developing agent, and the total saponin content of different origins was determined by spectrophotometry at 560 nm. Preparation of the test solution: Take about 1.0g of the crude powder of Polygalaceae, accurately weighed, place it in the Soxhlet extractor, add the appropriate amount of ether, reflux for 4h, take out, remove the ether, add 40mL of methanol, and soak overnight. Add methanol to the appropriate amount, reflux for 4 h, extract the methanol to 10 mL, add 4 times the amount of diethyl ether, leave overnight, filter, and settle the precipitate to a volume of 25 mL with methanol as the test solution.
